This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Phospholipid Transfer Protein (PLTP) in serum, plasma, tissue homogenates or other biological fluids. The kit employs a pre-coated microplate format with biotinylated detection antibody and HRP-avidin conjugate chemistry, producing a colourmetric signal read at 450 nm in a microplate reader. Total assay time is 4 hours 30 minutes.

In the workflow, standards and test samples are added to antibody-coated wells where PLTP is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-PLTP Antibody binds the captured PLTP and an HRP-Avidin conjugate is added. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of PLTP captured, and concentration is calculated by reference to a standard curve.

The assay demonstrates an assay range of 0.625–40 ng/ml with a sensitivity of <0.247 ng/ml. Sample recovery has been validated in serum (80%–102%, average 91%), EDTA plasma (81%–100%, average 90%) and heparin plasma (80%–89%, average 84%) across five samples per matrix. Dilution linearity has been confirmed at 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or –20°C as instructed, and the kit must not be used past its expiration date.
