Human Phospholipase B ELISA Kit

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit intended for the in vitro quantitative determination of human Phospholipase B in tissue homogenates or other biological fluids. The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with the signal read on a colorimetric microplate reader at 450 nm. An antibody specific for Phospholipase is immobilised on the plate; standards and samples are added, followed by a Biotinylated Anti-Phospholipase Antibody and an HRP-Avidin conjugate. The HRP enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow upon addition of the acidic stop solution, and the absorbance at 450 nm is proportional to the amount of Phospholipase captured in each well.

The reported assay range is 0.312-20 ng/ml and the sensitivity is stated as <0.113 ng/ml, with a total assay time of 4h 30m. Recovery data supplied with the kit indicate average values of 91% for serum (range 80%-102%, n=5), 90% for EDTA plasma (range 81%-100%, n=5) and 84% for Heparin plasma (range 80%-89%, n=5). Dilution linearity from 1:2 to 1:16 is reported across serum, EDTA plasma and Heparin plasma samples.

Kit components include the pre-coated 96 well microplate, a lyophilised standard, 120 µl each of Detection Solution A and Detection Solution B, 20 ml of 30X Wash Buffer, 45 ml of Sample Dilution Buffer, 9 ml of TMB Substrate, 6 ml of Stop Solution and adhesive plate sealers.
