Human Phospholipase A2, Calcium Independent ELISA Kit

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human Calcium-Independent Phospholipase A2/PLA2G6 in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) with a colourimetric readout on a standard microplate reader. The workflow involves an immobilised capture antibody binding target from standards or test samples, followed by a biotinylated anti-PLA2G6 detection antibody, HRP-avidin conjugate, and TMB substrate producing a yellow product proportional to analyte concentration, measured at 450 nm.

The working range is 78.1–5000 pg/ml with a sensitivity of <32 pg/ml. Recovery has been evaluated in serum (average 91%, range 80–102%, n=5), EDTA plasma (average 90%, range 81–100%, n=5), and heparin plasma (average 84%, range 80–89%, n=5). Dilution linearity has been demonstrated across 1:2, 1:4, 1:8, and 1:16 dilutions in serum, EDTA plasma, and heparin plasma (n=5 per matrix). Total assay time is 4 h 30 min. Kit components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
