Human Phosphoinositide-3-Kinase Catalytic Delta Polypeptide ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human PI 3 Kinase p110 delta in tissue homogenates, cell lysates and other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for PI 3 Kinase p110 delta pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and any PI 3 Kinase p110 delta present is captured by the immobilised antibody. After washing, a Biotinylated Anti-PI 3 Kinase p110 delta Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated by reference to a standard curve.

The assay has a reported product range of 0.312–20 ng/ml and a sensitivity of less than 0.086 ng/ml, with a total assay time of 4 hours 30 minutes. Sample recovery has been characterised in serum (80%–102%, average 91%), EDTA plasma (81%–101%, average 91%) and heparin plasma (80%–89%, average 84%). Dilution linearity has been validated across 1:2, 1:4, 1:8 and 1:16 dilutions in the same three sample types. Kit components should be stored at +4°C or –20°C as instructed, and the kit must not be used past its expiration date.
