This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com is designed for the in vitro quantitative determination of human phosphodiesterase 10A (PDE10A) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using a microplate pre-coated with an antibody specific for PDE10A. Standards and test samples are added to the wells and the PDE10A present in each sample is bound by the immobilized antibody. After incubation and washing, the wells are incubated with a biotinylated anti-PDE10A antibody, followed by an HRP-avidin conjugate. A TMB substrate solution is then used to visualize the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of PDE10A captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a reported sensitivity of <0.104 ng/ml and a standard curve range of 0.312-20 ng/ml. The platform is a pre-coated 96-well microplate configured as 12 x 8 well strips, with a total assay time of approximately 4 hours 30 minutes. Detection is colourimetric. The kit is for use with human samples. Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solution A, detection solution B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers; components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Recovery and dilution linearity have been characterised for serum, EDTA plasma, and heparin plasma samples, with reported average recoveries of 91% (serum, n=5, range 83%-100%), 97% (EDTA plasma, n=5, range 91%-103%), and 95% (heparin plasma, n=5, range 89%-102%). Dilution linearity has also been reported across 1:2, 1:4, 1:8, and 1:16 dilutions for these matrices.
