Antibodies.com supplies this sandwich ELISA for the in vitro quantitative determination of human RKIP (also referred to as Phosphatidylethanolamine Binding Protein 1) in tissue homogenates, cell lysates, or other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips and follows a standard sandwich enzyme immunoassay workflow: a capture antibody specific for RKIP is immobilised on the plate, standards and samples are added and bound by the immobilised antibody, the wells are washed and incubated with a Biotinylated Anti-RKIP Antibody, and an HRP-Avidin conjugate is then added. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow on addition of acidic stop solution; the density of yellow is proportional to the amount of RKIP captured in each well and is measured by absorbance at 450 nm in a colorimetric microplate reader.

According to the manufacturer, the kit has a sensitivity of <0.058 ng/ml and a quantitative range of 0.156–10 ng/ml, with a total assay time of 4 h 30 min. Reported average recovery (n=5) is 91% (range 80%–102%) in serum, 90% (range 81%–100%) in EDTA plasma, and 84% (range 80%–89%) in heparin plasma. The kit includes the pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, and components are stored at +4°C or –20°C as instructed. It should not be used past the stated expiration date.
