Human Phosphatase And Tensin Homolog ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human PTEN (Phosphatase And Tensin Homolog) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for PTEN immobilised on the plate. Standards and test samples are added to the wells, followed by a Biotinylated Anti-PTEN Antibody and an HRP-Avidin conjugate. After incubation and washing, TMB substrate solution is added, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow, read at 450 nm on a colorimetric microplate reader, is proportional to the amount of PTEN captured in each well, and the concentration is calculated by reference to a standard curve.

Reactivity is for human samples, with a stated sensitivity of <0.136 ng/ml and a standard curve range of 0.312–20 ng/ml. The total assay time is 4h 30m. Average recovery values reported by the manufacturer are 80%–102% for serum, 81%–100% for EDTA plasma, and 80%–89% for heparin plasma.
