Human Peroxisome Proliferator Activated Receptor Delta ELISA Kit

An Antibodies.com product, this sandwich ELISA enables the in vitro quantitative determination of human PPAR delta (Peroxisome Proliferator Activated Receptor Delta) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a pre-coated 96-well microplate to which an antibody specific for PPAR delta has been immobilised. Standards and test samples are added to the wells, followed by a Biotinylated Anti-PPAR delta Antibody and then an HRP-Avidin conjugate. A TMB substrate is added to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of the stop solution; absorbance is read at 450 nm in a microplate reader and PPAR delta concentration is calculated from the standard curve.

The assay has a working range of 0.156–10 ng/ml and a sensitivity of 0.057 ng/ml, with a total assay time of 4h 30m. Reported average recoveries (n=5) span 80–102% for serum (mean 91%), 81–100% for EDTA plasma (mean 90%) and 80–89% for heparin plasma (mean 84%). Dilution linearity has been characterised at 1:2, 1:4, 1:8 and 1:16 in each of serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date. Detection is colorimetric and is read on a standard microplate reader.
