Human Peroxisome Proliferator Activated Receptor Alpha ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Peroxisome Proliferator Activated Receptor Alpha (PPAR alpha) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a 96-well microplate pre-coated with an antibody specific for PPAR alpha, and is read on a colorimetric microplate reader at 450 nm.

The working range covers 0.156-10 ng/ml with a sensitivity of 0.052 ng/ml, and the assay can be completed in 4 hours 30 minutes. Standards and test samples are incubated in the pre-coated wells and bound by the immobilised antibody, followed by incubation with a Biotinylated Anti-PPAR alpha Antibody and then an HRP-Avidin conjugate. The enzymatic reaction is visualised using a TMB substrate that produces a blue colour, changing to yellow upon addition of the stop solution; the intensity of the yellow colour is proportional to the amount of PPAR alpha captured in each well, allowing quantification against a standard curve.

Recovery has been demonstrated in serum, EDTA plasma and heparin plasma, with average values of 91% (range 80%-102%) in serum, 90% (range 81%-100%) in EDTA plasma and 84% (range 80%-89%) in heparin plasma.
