Description
From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Peroxiredoxin 6 in serum, plasma, tissue homogenates or other biological fluids. The 96-well microplate is pre-coated with a capture antibody specific for Peroxiredoxin 6. Standards and samples are added to the wells; following incubation and washing, a Biotinylated Anti-Peroxiredoxin 6 Detection Antibody and an HRP-Avidin conjugate are applied sequentially. TMB substrate is then added, producing a blue colour that changes to yellow upon addition of an acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader.
Quantitative performance includes a sensitivity of 0.108 ng/ml and an assay range of 0.312–20 ng/ml. Reported spike-and-recovery values are: serum (n=5) 80%–102% (average 91%); EDTA plasma (n=5) 81%–100% (average 90%); heparin plasma (n=5) 80%–89% (average 84%). Linearity has been evaluated across 1:2, 1:4, 1:8 and 1:16 dilutions for each matrix. Total assay time is 4 hours 30 minutes and detection is colorimetric.
Kit components supplied include a pre-coated 96-well microplate (12 × 8 well strips), lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or –20°C as instructed; the kit should not be used past its expiration date.
