Human Perforin 1 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Perforin in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated microplate onto which an antibody specific for Perforin has been immobilised. Standards and test samples are added to the wells, and captured Perforin is detected using a biotinylated anti-Perforin antibody followed by an HRP-avidin conjugate. Signal is visualised with TMB substrate, producing a yellow product after addition of stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of Perforin is calculated from the standard curve.

The stated sensitivity is less than 0.56 ng/ml and the working range is 1.56–100 ng/ml. The total assay time is 4 h 30 min. Recovery data (n=5) report averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Reported dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions shows ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (each n=5).
