Human Peptide YY ELISA Kit

This Antibodies.com competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Peptide YY/PYY in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) and the competitive enzyme immunoassay technique, in which analyte present in the sample or standard competes with immobilised Peptide YY/PYY for binding sites on a biotinylated anti-Peptide YY/PYY antibody. Unbound antibody is removed by washing, an HRP-Avidin conjugate is added, and TMB substrate is used to visualise the reaction; the resulting yellow colour density is inversely proportional to the amount of Peptide YY/PYY, with concentration calculated from the O.D. absorbance at 450 nm against a standard curve.

The kit has a stated sensitivity of <0.48 pg/ml, a working range of 1.56–100 pg/ml, and a total assay time of 2h 30m. Detection is colourimetric, read on a microplate reader. Component storage is +4 °C or -20 °C as instructed, and the kit should not be used past its expiration date.

Reported recovery from undiluted samples: Serum (n=5) range 80%–102% (average 91%), EDTA Plasma (n=5) 81%–100% (average 90%), Heparin Plasma (n=5) 80%–89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions: Serum (n=5) 87–91%, 87–107%, 74–101%, 92–97%; EDTA Plasma (n=5) 90–105%, 84–101%, 90–101%, 79–108%; Heparin Plasma (n=5) 84–95%, 92–105%, 82–105%, 89–91%.
