Human Peptide YY 3 ELISA Kit

Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Peptide YY 3 (PYY3) in serum, plasma or other biological fluids. The assay employs a competitive enzyme immunoassay technique on a pre-coated microplate arranged as 12 x 8 well strips. During incubation, PYY3 in samples or standards competes with the fixed amount of immobilized PYY3 for binding sites on a Biotinylated Anti-PYY3 Antibody. After washing, an HRP-Avidin conjugate is added followed by TMB substrate solution; the resulting colourmetric signal is read at 450 nm and is inversely proportional to the amount of PYY3 present in the sample or standard.

Reported performance specifications include an assay range of 15.6–1000 pg/ml, a sensitivity of 4.12 pg/ml, and a total assay time of approximately 2 hours 30 minutes. The assay is validated for use with human samples. Recovery data show ranges of 80%–102% (average 91%) for serum, 81%–100% (average 90%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma. Dilution linearity across 1:2 to 1:16 dilutions (n=5) yielded 87–91% to 92–97% for serum, 90–105% to 79–108% for EDTA plasma, and 84–95% to 89–91% for heparin plasma.

Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or −20°C as indicated, and the kit must not be used past its expiration date. A colorimetric microplate reader is required for measurement.
