From Antibodies.com, this sandwich ELISA is intended for the in vitro quantitative determination of human PNPLA3 in tissue homogenates, cell lysates or other biological fluids, with reactivity confirmed for human samples.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for PNPLA3 is immobilised on the plate; standards and samples are added, then, after incubation and washing, a Biotinylated Anti-PNPLA3 Antibody followed by an HRP-Avidin conjugate are applied. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. PNPLA3 concentration is calculated against a standard curve. Reported sensitivity is <0.29 ng/ml, the measuring range is 0.781-50 ng/ml, and total assay time is 4h 30m.

Recovery (n=5) averaged 91% in serum (range 80%–102%), 91% in EDTA plasma (81%–101%) and 84% in heparin plasma (80%–89%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma.
