A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Parvalbumin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.
The assay uses a pre-coated 96-well microtiter plate formatted as 12 x 8 well strips, with an antibody specific for Parvalbumin immobilised on the wells. Standards and samples are added, followed by a biotinylated anti-Parvalbumin antibody and an HRP-Avidin conjugate. A TMB substrate is then added; catalysis produces a blue product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450nm. The reported product range is 1.56-100 ng/ml with a sensitivity of 0.55 ng/ml, and the total assay time is 4h 30m.
Reported recovery ranges are 80%-102% for serum (n=5), 81%-100% for EDTA plasma (n=5), and 80%-89% for heparin plasma (n=5). Linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 dilutions for the same sample types. Kit components are stored at +4°C or -20°C, and detection is performed on a colorimetric microplate reader.
