Description

From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Parathyroid Hormone in serum, plasma or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with Parathyroid Hormone antigen. During incubation, Parathyroid Hormone in samples or standards competes with the immobilized antigen for binding sites on a Biotinylated Anti-Parathyroid Hormone Antibody; the more analyte present, the less antibody binds to the plate. After washing, an HRP-Avidin conjugate is added, and TMB substrate is used to visualize the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of yellow colour is inversely proportional to Parathyroid Hormone concentration, calculated by reading absorbance at 450 nm against a standard curve.

The kit has a reported product range of 1.56–100 pg/ml and a sensitivity of 0.49 pg/ml. Detection is colorimetric on a pre-coated microplate comprising 12 x 8 well strips, read with a colorimetric microplate reader, with a total assay time of approximately 2 hours 30 minutes. Reported recovery ranges across n=5 samples are: serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%), and heparin plasma 80%–89% (average 84%). Dilution linearity (1:2 to 1:16) was reported as 87–91% to 92–97% for serum, 90–105% to 79–108% for EDTA plasma, and 84–95% to 89–91% for heparin plasma. Kit components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
