Human Pantothenate Kinase 4 ELISA Kit

From Antibodies.com, this sandwich ELISA (sELISA) is designed for the in vitro quantitative determination of human Pantothenate Kinase 4 in tissue homogenates or other biological fluids. The kit uses a 96-well microtiter plate pre-coated with an antibody specific for Pantothenate Kinase 4. Standards and test samples are added to the wells and the Pantothenate Kinase 4 present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Pantothenate Kinase 4 Antibody is added and binds the captured target, after which an HRP-Avidin conjugate is added. TMB substrate is then used to visualise the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of Pantothenate Kinase 4 captured in each well, and concentration is calculated by reading absorbance at 450 nm in a microplate reader and referring to the standard curve.

Reported sensitivity is 0.064 ng/ml (also cited as <0.064 ng/ml) and the assay range is 0.156–10 ng/ml. Total assay time is 4 hours 30 minutes. Detection is colourimetric, read on a colorimetric microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
