This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Pancreatic Polypeptide in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips, with colourimetric detection read at 450 nm on a microplate reader.

During incubation, Pancreatic Polypeptide present in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-Pancreatic Polypeptide Antibody. After washing, an HRP-Avidin conjugate is added, and TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow colour is inversely proportional to the amount of Pancreatic Polypeptide, and concentration is calculated by reference to a standard curve. The stated product range is 46.8-3000 pg/ml with a sensitivity of 13.3 pg/ml, and the assay time is 2h 30m. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.

Manufacturer validation data report recovery in serum (range 80% - 102%, average 91%), EDTA plasma (range 81% - 100%, average 90%) and heparin plasma (range 80% - 89%, average 84%), together with dilution linearity values for 1:2, 1:4, 1:8 and 1:16 dilutions in each of these sample types.
