Human Palladin ELISA Kit

Antibodies.com supplies this sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human Palladin in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate with an antibody specific for Palladin, where standards and samples are added and Palladin is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Palladin Antibody is added, then HRP-Avidin conjugate, with visualisation via TMB substrate producing a blue-coloured product that turns yellow after addition of acidic stop solution.

The density of yellow is proportional to the amount of Palladin captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader against the standard curve. The stated assay range is 0.156–10 ng/ml with a sensitivity of <0.051 ng/ml, and the total assay time is 4h 30m. Recovery performance is reported for serum (n=5) at 80%–102% (average 91%), EDTA plasma (n=5) at 81%–100% (average 90%), and heparin plasma (n=5) at 80%–89% (average 84%). Linearity of dilution has been assessed for serum, EDTA plasma, and heparin plasma at 1:2, 1:4, 1:8, and 1:16 dilutions.
