Human p53 ELISA Kit

From Antibodies.com, this sandwich ELISA (sELISA) is intended for the in vitro quantitative determination of human p53 in serum, plasma, cell lysates, tissue homogenates, and other biological fluids. Reported sensitivity is < 10 pg/ml with an assay range of 156-10,000 pg/ml, and the kit is stated to be specific for p53 with no reported cross-reactivity against other analogs or variants. The 4-hour protocol uses a pre-coated microplate with colorimetric detection read at 450 nm, and the kit is shipped at +4°C, with further storage at +4°C or -20°C as detailed in the manual; the expiry date and freeze/thaw restrictions noted in the manual should be observed.

Serum and plasma samples should be diluted at least 1:2 with Sample Dilution Buffer before testing; validated sample dilutions include healthy serum (1:2) and HeLa cell lysate (1:2-1:5). In the workflow, standards and samples are added to wells where p53 binds the immobilised capture antibody; after washing, a biotinylated anti-p53 detection antibody is added to form an antibody-antigen-antibody sandwich, followed by HRP-Streptavidin conjugate and TMB substrate. The resulting yellow product is read at 450 nm and the p53 concentration is calculated against the standard curve.
