Human Oxoglutarate Dehydrogenase ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Oxoglutarate Dehydrogenase (OGDH) in tissue homogenates, cell lysates or other biological fluids. The kit employs a 96-well microplate pre-coated with an antibody specific for OGDH. Standards and test samples are added to the wells, where OGDH binds to the immobilized antibody. After incubation and washing, a Biotinylated Anti-OGDH Antibody is added, binding the captured OGDH in each well. An HRP-Avidin conjugate is then added, and following further incubation and washing, TMB substrate solution is used to visualize the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow colour is proportional to the amount of OGDH captured, and concentration is calculated by reading absorbance at 450 nm against a standard curve.

The kit has a reported sensitivity of <0.59 ng/ml and an assay range of 1.56-100 ng/ml. The assay is run on a colorimetric microplate reader with a total assay time of approximately 4 hours 30 minutes. Recovery performance has been evaluated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5) and heparin plasma (range 80%-89%, average 84%, n=5). Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
