Detection uses a sandwich ELISA workflow on a pre-coated microplate. A biotinylated detection antibody binds the captured analyte to form an antibody–antigen–antibody sandwich complex, after which HRP-Streptavidin conjugate and TMB substrate are added. The absorbance of the resulting coloured product is read at 450 nm, with intensity directly proportional to Osteopontin concentration and quantified against the standard curve.
