Human Ornithine Decarboxylase Antizyme 1 ELISA Kit

Antibodies.com offers a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human Ornithine Decarboxylase Antizyme 1 (OAZ1) in tissue homogenates or other biological fluids. The assay employs a pre-coated microplate with an antibody specific for OAZ1. Standards and test samples are added to the wells and bound by the immobilised antibody, followed by incubation with a biotinylated anti-OAZ1 detection antibody and HRP-avidin conjugate. After washing, TMB substrate is added; the enzymatic reaction produces a blue product that turns yellow upon addition of an acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and OAZ1 concentration is calculated against a standard curve.

Reported performance characteristics include a sensitivity of <0.058 ng/ml, an assay range of 0.156–10 ng/ml, and a total assay time of 4 hours 30 minutes. Spike-and-recovery validation in serum (n=5) showed a range of 80%-102% and an average of 91%; EDTA plasma (n=5) showed a range of 81%-100% and an average of 90%; heparin plasma (n=5) showed a range of 80%-89% and an average of 84%. Dilutional linearity was assessed at 1:2, 1:4, 1:8, and 1:16 in serum, EDTA plasma, and heparin plasma samples.
