Human Ornithine Carbamoyl Transferase ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Ornithine Carbamoyltransferase/OTC in serum, plasma, tissue homogenates or other biological fluids. The assay uses a microtiter plate pre-coated with an antibody specific for Ornithine Carbamoyltransferase/OTC. Standards and test samples are added to the wells, followed by a Biotinylated Anti-Ornithine Carbamoyltransferase/OTC Antibody and then an HRP-Avidin conjugate. TMB substrate is used to visualise the reaction, producing a blue product that changes to yellow on addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of Ornithine Carbamoyltransferase/OTC captured in each well and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against the standard curve.

The assay has a product range of 0.468–30 ng/ml and a sensitivity of 0.16 ng/ml. The platform is a pre-coated microplate (12 x 8 well strips) and the total assay time is 4h 30m. Detection is colorimetric and kit components are stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date. Reported average recovery values were 91% for serum (n=5, range 80%–102%), 90% for EDTA plasma (n=5, range 81%–100%) and 84% for heparin plasma (n=5, range 80%–89%). Across sample dilutions from 1:2 to 1:16, the reported linearity was 74%–107% for serum (n=5), 79%–108% for EDTA plasma (n=5) and 82%–105% for heparin plasma (n=5).
