Human Orexin ELISA Kit

This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Orexin in serum, plasma, tissue homogenates or other biological fluids. The assay employs the competitive enzyme immunoassay technique, in which Orexin present in samples or standards competes with Orexin antigen pre-coated on the plate for binding sites on a biotinylated anti-Orexin antibody. Detection uses an HRP-avidin conjugate and TMB substrate, with absorbance read at 450 nm on a colorimetric microplate reader; the resulting yellow colour intensity is inversely proportional to the Orexin concentration in the sample.

The kit provides a quantitative measuring range of 15.6–1000 pg/ml with a sensitivity of 4.22 pg/ml and a total assay time of approximately 2 hours 30 minutes. Kit components should be stored at +4 °C or –20 °C as instructed, and the kit must not be used past its expiration date.

Validation data demonstrate consistent recovery across sample matrices. In undiluted samples (n=5), average recoveries were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–99%), and 84% for heparin plasma (range 80–89%). Linearity was maintained across dilutions from 1:2 through 1:16 in serum, EDTA plasma and heparin plasma, supporting reliable quantitation across biological fluids.
