A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is intended for the in vitro quantitative determination of human Nucleophosmin in tissue homogenates, cell lysates and other biological fluids. The kit is reactive with human samples and is run on a pre-coated microplate platform configured as 12 × 8-well strips.
Quantitative performance is defined by an assay range of 0.312–20 ng/ml and a sensitivity of 0.128 ng/ml. The full protocol requires 4 h 30 m of incubation, with absorbance read at 450 nm on a colorimetric microplate reader. Components are stored at +4 °C or −20 °C as directed on the individual labels, and the kit should not be used past its expiration date. Detection uses a biotinylated anti-Nucleophosmin antibody together with an HRP-avidin conjugate and TMB substrate, producing a yellow colour whose intensity is proportional to the amount of Nucleophosmin captured per well.
Reported recovery values across n=5 are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity is also documented at 1:2, 1:4, 1:8 and 1:16 across the same three matrices, with per-dilution ranges given in the product documentation.
