Human Nuclear Receptor Related Protein 1 ELISA Kit

From Antibodies.com, this sandwich ELISA enables the in vitro quantitative determination of human Nuclear Receptor Related Protein 1 (Nurr1) in tissue homogenates, cell lysates and other biological fluids. A Nurr1-specific antibody is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added, captured Nurr1 is bound by a Biotinylated Anti-Nurr1 Antibody, and detection is achieved with HRP-Avidin conjugate and TMB substrate. The reaction yields a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the Nurr1 concentration is calculated against the standard curve.

According to the manufacturer, the assay range is 0.156-10 ng/ml with a sensitivity of <0.053 ng/ml, and the total assay time is 4h 30m. Reported mean recoveries (n=5) are 91% for serum (range 80%-102%), 90% for EDTA plasma (81%-100%) and 84% for heparin plasma (80%-89%). Reported linearity of dilution (n=5) at 1:2, 1:4, 1:8 and 1:16 is 87-91%, 87-107%, 74-101% and 92-97% respectively for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.

Kit components comprise a lyophilised standard (2 vials), Detection Solution A (120 μl) and Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers. Components are stored between +4°C and -20°C as instructed, and the kit should not be used past its expiration date.
