Human Nitric Oxide Synthase 1, Neuronal ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human nNOS (neuronal) — also known as Nitric Oxide Synthase 1, Neuronal — in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microtiter plate (12 x 8 well strips) bearing a capture antibody specific for the neuronal isoform of nitric oxide synthase. Standards and samples are added to the wells; following incubation and washing, a biotinylated anti-nNOS (neuronal) detection antibody binds the captured analyte, and HRP-Avidin conjugate is then added. The enzymatic reaction is visualised with TMB substrate, which produces a blue colour that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the nNOS (neuronal) concentration is calculated against the standard curve.

Reported performance figures include an assay range of 0.156–10 ng/ml and a sensitivity of 0.053 ng/ml. Spike-and-recovery experiments gave ranges of 80–102% (serum, n=5, average 91%), 81–100% (EDTA plasma, n=5, average 90%) and 80–89% (heparin plasma, n=5, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Total assay time is 4 hours 30 minutes and detection is colourimetric.
