From Antibodies.com, this Human Niemann Pick Disease Type C2 ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Niemann Pick C2 in serum, plasma, tissue homogenates, or other biological fluids. The assay uses a pre-coated microplate together with a biotinylated anti-Niemann Pick C2 antibody, an HRP-Avidin conjugate and TMB substrate; the resulting yellow product is read at 450 nm and quantified against a standard curve.

Quoted performance characteristics are a sensitivity of <0.31 ng/ml, an assay range of 0.781–50 ng/ml and a total assay time of 4h 30m. Recovery was assessed with n=5 samples per matrix: serum ranged from 80% to 102% (average 91%), EDTA plasma from 81% to 100% (average 90%), and heparin plasma from 80% to 89% (average 84%). Linearity of dilution was also evaluated across n=5 samples at 1:2, 1:4, 1:8 and 1:16, giving ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.

The kit is supplied with a pre-coated 96-well microplate, lyophilised standard, two detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
