The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips and follows the competitive enzyme immunoassay technique. Neuropeptide Y in the samples or standards competes with immobilised Neuropeptide Y antigen for binding sites on a Biotinylated Anti-Neuropeptide Y Antibody. After washing, an HRP-Avidin conjugate is added, and TMB substrate visualises the reaction by producing a blue product that turns yellow upon addition of acidic stop solution; the resulting yellow intensity is inversely proportional to the Neuropeptide Y concentration. Absorbance is read at 450 nm in a colorimetric microplate reader and quantified against a standard curve. The stated product range is 3.12-200 pg/ml with sensitivity of <0.97 pg/ml, and total assay time is 2h 30m. Reported recovery averages are 91% for serum, 90% for EDTA plasma and 84% for heparin plasma, with linearity demonstrated across serial dilutions.
