Human Neuronal Pentraxin II ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of human Neuronal Pentraxin II (NPTX2) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microplate format organised as 12 x 8 well strips, with a colorimetric readout at 450 nm. Captured NPTX2 is detected using a biotinylated anti-NPTX2 antibody followed by an HRP-avidin conjugate, with TMB substrate producing a blue product that turns yellow upon addition of acidic stop solution.

Reported sensitivity is 0.064 ng/ml and the assay range is 0.156-10 ng/ml. Total assay time is 4h 30m. Recovery performance in neat samples (n=5) averaged 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%), and 84% in heparin plasma (range 80%–89%). Linearity across serial dilutions of 1:2, 1:4, 1:8 and 1:16 was assessed in serum (n=5; ranges 87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (n=5; ranges 90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (n=5; ranges 84–95%, 92–105%, 82–105%, 89–91%). Standards are prepared as a 1:2 serial dilution from a 10 ng/ml stock down to 0.156 ng/ml.

Kit components include the pre-coated microplate, two vials of lyophilized standard, detection solutions A and B (120 μl each), a 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml), and adhesive plate sealers.
