Human Neuromedin U ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Neuromedin U (NMU) in serum, plasma or other biological fluids.

The assay employs a pre-coated microplate to which an NMU-specific antibody is immobilised. Standards and test samples are added to the wells and any NMU present is bound by the immobilised antibody. Following incubation and washing, a biotinylated anti-NMU antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction; the resulting blue product turns yellow upon addition of acidic stop solution, and the intensity of yellow—read as absorbance at 450 nm in a colorimetric microplate reader—is proportional to the NMU concentration, which can be calculated by reference to a standard curve.

The kit offers a sensitivity of <0.059 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. In undiluted samples (n=5), average recovery was 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Linearity assessed at sample dilutions of 1:2, 1:4, 1:8 and 1:16 produced recoveries of 74–107% in serum, 79–108% in EDTA plasma and 82–105% in heparin plasma.
