Human Neurogranin ELISA Kit

Designed for the in vitro quantitative determination of human Neurogranin in tissue homogenates, cell lysates, or other biological fluids, this sandwich ELISA kit from Antibodies.com is configured on a pre-coated 96-well microplate arranged as 12 x 8 well strips and is read on a colorimetric microplate reader. The total assay time is 4 hours 30 minutes, and the kit should not be used past its stated expiration date.

The assay uses a sandwich immunoassay format in which an antibody specific for Neurogranin is pre-coated onto the plate. Standards and test samples are added to the wells, allowing Neurogranin present in each sample to be captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Neurogranin Antibody is added, which binds the captured Neurogranin, followed by an HRP-Avidin conjugate. TMB substrate is then introduced; HRP catalysis produces a blue-coloured product that changes to yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm, and the density of yellow is proportional to the amount of Neurogranin captured in each well, with concentration calculated by reference to the standard curve.

Quoted performance figures include a sensitivity of <0.044 ng/ml and an assay range of 0.156-10 ng/ml. Recovery was assessed in serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%), and heparin plasma (n=5, range 80%-89%, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was demonstrated for serum (87%-91%, 87%-107%, 74%-101%, 92%-97%), EDTA plasma (90%-105%, 84%-101%, 90%-101%, 79%-108%) and heparin plasma (84%-95%, 92%-105%, 82%-105%, 89%-91%).
