Human Neuroglobin ELISA Kit

A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human Neuroglobin in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Neuroglobin that is pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where Neuroglobin is captured by the immobilized antibody. After washing, a Biotinylated Anti-Neuroglobin Antibody binds the captured target, followed by incubation with HRP-Avidin conjugate. Following further washing, a TMB substrate solution is used to visualize the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The concentration of Neuroglobin is calculated from the absorbance read at 450 nm in a microplate reader, using the standard curve.

The kit shows a sensitivity of <0.052 ng/ml and an assay range of 0.156–10 ng/ml. Detection is colorimetric, read on a microplate reader, and the total assay time is 4 hours 30 minutes. Spike-and-recovery data are provided: serum samples (n=5) gave an average of 91% (range 80–102%), EDTA plasma (n=5) gave 90% (range 81–100%), and heparin plasma (n=5) gave 84% (range 80–89%). Dilution linearity data at 1:2, 1:4, 1:8 and 1:16 dilutions are also reported for serum, EDTA plasma and heparin plasma samples. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
