A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) designed for the in vitro quantitative determination of human NET1 (Neuroepithelial Cell Transforming Gene 1) in tissue homogenates or other biological fluids. Marketed by Antibodies.com, the kit uses a pre-coated microplate whose wells are bound by an antibody specific for NET1. Standards and test samples are added, followed by incubation with a Biotinylated Anti-NET1 Antibody and an HRP-Avidin conjugate. A TMB substrate solution visualises the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450 nm on a microplate reader and the NET1 concentration calculated by reference to the standard curve.

The assay has a stated sensitivity of 0.125 ng/ml and an assay range of 0.312-20 ng/ml, with a total assay time of 4 hours 30 minutes. Detection is colourimetric on a microplate reader. Recovery has been characterised in serum, EDTA plasma and heparin plasma, with average recoveries of 91%, 90% and 84% respectively across 5 samples per matrix. Kit components include a pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at either +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
