Human Nestin ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Nestin in tissue homogenates, cell lysates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for Nestin. Standards and test samples are added to the wells, where Nestin binds to the immobilized capture antibody. After washing, a biotinylated anti-Nestin antibody is added, followed by HRP-Avidin conjugate and TMB substrate solution. The enzymatic reaction produces a blue product that changes to yellow upon addition of acidic stop solution, with colour intensity proportional to Nestin concentration. Absorbance is read at 450 nm in a colorimetric microplate reader and compared against a standard curve.

The kit demonstrates a sensitivity of <0.065 ng/ml with a working range of 0.156–10 ng/ml, and a total assay time of 4 hours 30 minutes. Reported recovery rates in spiked samples average 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–100%), and 84% for heparin plasma (n=5, range 80–89%). Components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
