Human Neprilysin ELISA Kit

This Sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Neprilysin in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a 96-well microplate pre-coated with an antibody specific for CD10. Standards and test samples are added to the wells, where CD10 is captured by the immobilised antibody. After washing, a Biotinylated Anti-CD10 Antibody binds the captured target. An HRP-Avidin conjugate is then added, and TMB substrate is used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of analyte captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

Components supplied include a pre-coated microplate configured as 12 x 8 well strips, lyophilised standard vials, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Total assay time is 4 hours 30 minutes. Reported sensitivity is <5.8 pg/ml, with an assay range of 15.6-1000 pg/ml. Average recovery values are reported as 91% for serum (n=5, range 80-102%), 90% for EDTA plasma (n=5, range 81-100%), and 84% for heparin plasma (n=5, range 80-89%). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma. Components should be stored at +4°C or -20°C as indicated, and the kit is suitable for use on a colourmetric microplate reader platform.
