Human NADH Dehydrogenase, Quinone 1 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com for the in vitro quantitative determination of human NQO1 (NADH Dehydrogenase, Quinone 1) in tissue homogenates, cell lysates and other biological fluids. The assay employs a 96-well microplate pre-coated with an anti-NQO1 capture antibody; standards and samples are added to the wells, washed, and incubated sequentially with a biotinylated anti-NQO1 detection antibody and an HRP-avidin conjugate. TMB substrate is then added to visualise the HRP reaction as a blue product that turns yellow after addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader and NQO1 concentration calculated against the standard curve.

Reported sensitivity is 0.124 ng/ml and the working assay range is 0.312–20 ng/ml. Total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.

Spike-and-recovery validation in serum (n=5) gave a range of 80–102% (mean 91%); EDTA plasma (n=5) 81–100% (mean 90%); heparin plasma (n=5) 80–89% (mean 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 produced recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (all n=5).
