Configured to quantify human Na-K-Cl Cotransporter 2 (gene symbol SLC12A1, protein alias NKCC2), this Antibodies.com kit targets the apical-membrane transporter that drives electroneutral Na⁺, K⁺, and 2 Cl⁻ uptake in the thick ascending limb of the loop of Henle. The assay is intended for research use only in studies of renal salt handling, furosemide and other loop diuretic pharmacology, and the molecular pathology of Bartter syndrome type I.

The 96-test configuration includes a pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, lyophilized recombinant standard, dilution buffer, wash concentrate, and TMB substrate. Each well is loaded with 100 µL of prepared sample drawn from cell lysates, kidney cortex homogenates, or transfected cell-line extracts. Reconstituted standards and any unused diluted antibody should be divided into single-use aliquots and frozen to limit repeated freeze-thaw cycles.

Capture and detection reagents are calibrated to the human SLC12A1 sequence and show no significant cross-reactivity with rat Slc12a1 or with the paralogous NKCC1 transporter encoded by SLC12A2, which is useful when distinguishing tubular NKCC2 activity from basolateral NKCC1 in renal preparations.
