Human Na-Cl Cotransporter ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human SLC12A3 (the Na-Cl Cotransporter) in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colorimetric detection performed by reading O.D. absorbance at 450 nm in a microplate reader. Reported sensitivity is 0.123 ng/ml with a standard range of 0.312-20 ng/ml, and total assay time is approximately 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed, and the product is for research use only, not intended for diagnostic or therapeutic use.

Sample recovery has been evaluated in serum (n=5, average 91%, range 80-102%), EDTA plasma (n=5, average 90%, range 81-100%) and heparin plasma (n=5, average 84%, range 80-89%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 is reported at 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. The capture antibody specific for SLC12A3 is immobilised on the plate, with a biotinylated anti-SLC12A3 detection antibody and HRP-Avidin conjugate, visualised using TMB substrate that yields a yellow product after acid stop solution is added.
