A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of human NT-ProBNP in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for NT-ProBNP pre-coated onto a microtiter plate. Standards and test samples are added and any NT-ProBNP present is captured by the immobilized antibody.

After washing, a Biotinylated Anti-NT-ProBNP Antibody is added, followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that changes to yellow after addition of an acidic stop solution, with the density of yellow proportional to the amount of NT-ProBNP captured in each well. Absorbance is read at 450 nm using a colorimetric microplate reader and the concentration of NT-ProBNP is calculated by reference to a standard curve.

The quantitative assay has a reported product range of 39-2500 pg/ml, a sensitivity of <15 pg/ml and a total assay time of 4h 30m. The kit is supplied with a pre-coated microplate and the necessary reagents, and is intended for research use only.
