Human N-Acylethanolamine Acid Amidase ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com is intended for the in vitro quantitative determination of human ASAHL/PLT in tissue homogenates, cell lysates or other biological fluids. The product has human reactivity and is supplied as a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for ASAHL/PLT is immobilised on the plate; standards and samples are added and any ASAHL/PLT present is captured. After washing, a biotinylated anti-ASAHL/PLT antibody is added, followed by HRP-avidin conjugate. The reaction is visualised with TMB substrate solution, producing a blue product that turns yellow on addition of stop solution; absorbance is read at 450 nm in a colorimetric microplate reader.

The standard curve covers an assay range of 0.312–20 ng/ml with a sensitivity of <0.122 ng/ml. The complete assay time is 4 hours 30 minutes. Kit components are stored at +4°C or −20°C as instructed, and the product must not be used past its expiration date.

Spike-and-recovery values (n=5) have been reported for serum (range 80–102%, average 91%), EDTA plasma (range 81–100%, average 90%) and heparin plasma (range 80–89%, average 84%). Dilutional linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%).
