Human N-Acetyl Alpha-D-Glucosaminidase ELISA Kit

A sandwich ELISA kit from Antibodies.com for the in vitro quantitative determination of human N-Acetyl Alpha-D-Glucosaminidase (NAGLU/NAG) in serum, plasma, tissue homogenates or other biological fluids. The assay is reactive with human samples and is listed under the Assays and Kits category.

The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for NAGLU/NAG is immobilised on the plate; standards and test samples are added, the plate is washed, and a biotinylated anti-NAGLU/NAG antibody is then applied. An HRP-Avidin conjugate and TMB substrate are added in turn, producing a blue colour that turns yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the NAGLU/NAG concentration is calculated by reference to a standard curve. Total assay time is approximately 4 hours 30 minutes.

Reported sensitivity is <0.125 ng/ml and the assay range is 0.312–20 ng/ml. Spike-and-recovery testing (n=5) gave averages of 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%) and 84% for heparin plasma (range 80%–89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 across the same sample types. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
