Human Myostatin ELISA Kit

From Antibodies.com, this human Myostatin ELISA Kit is a quantitative sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro measurement of human myostatin in serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids. The assay uses a pre-coated microplate to which an antibody specific for myostatin has been immobilised; standards and samples are added and myostatin is captured by the bound antibody. After washing, a biotinylated anti-myostatin detection antibody is added, followed by HRP-avidin conjugate. TMB substrate is used to visualise the HRP-catalysed reaction, producing a blue product that turns yellow after addition of the acidic stop solution; absorbance is read at 450 nm in a microplate reader and the myostatin concentration is calculated from the standard curve [S1][S3].

The assay covers an analytical range of 0.781–50 ng/ml with a sensitivity of <0.25 ng/ml [S1][S3]. The total procedure takes approximately 4 hours 30 minutes from sample addition to result [S1]. Mean recovery has been validated in serum (n=5) at 91% over 80%–102%, in EDTA plasma (n=5) at 90% over 81%–100%, and in heparin plasma (n=5) at 84% over 80%–89% [S1]. Linearity on dilution has also been demonstrated: serum (n=5) at 87–91% (1:2), 87–107% (1:4), 74–101% (1:8) and 92–97% (1:16); EDTA plasma at 90–105%, 84–101%, 90–101% and 79–108% across the same dilutions; and heparin plasma at 84–95%, 92–105%, 82–105% and 89–91% [S1].

Kit components include a pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers [S1][S3]. Components should be stored at +4 °C or –20 °C as indicated on individual labels and the kit should not be used past its expiration date [S1][S3].
