Absorbance is measured on a colorimetric microplate reader at 450 nm, with analyte concentration calculated against the supplied standard curve.

An antibody specific for MYH7B is pre-coated onto a 96-well microplate provided as 12 x 8 well strips. Captured analyte is detected using a biotinylated anti-MYH7B antibody followed by an HRP-avidin conjugate and visualised with TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution. The reported assay range is 0.156-10 ng/ml with a sensitivity of less than 0.069 ng/ml, and the total assay time is 4h 30m.

Spike-and-recovery data (n=5) are reported for serum (average 91%, range 83-100%), EDTA plasma (average 95%, range 91-100%) and heparin plasma (average 96%, range 91-101%), alongside dilution linearity at 1:2, 1:4, 1:8 and 1:16 for the same three matrices.
