A sandwich enzyme-linked immunosorbent assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of Slow Skeletal Myosin Heavy Chain / Myosin Heavy Chain 7, Cardiac Muscle, Beta in human serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay uses an antibody specific for the target pre-coated onto a microplate. Standards and test samples are added to the wells, followed by a biotinylated anti-target antibody and an HRP-avidin conjugate. After incubation and washing, TMB substrate is added; catalysis produces a blue-coloured product that changes to yellow after addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the target concentration is calculated against the standard curve.

The format is a quantitative, colorimetric sandwich ELISA with reactivity against human samples. Reported performance figures include a measuring range of 0.156–10 ng/ml, a sensitivity of 0.053 ng/ml and an assay time of 4h 30m. Reported mean recoveries (n=5) were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was reported as 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components are stored at +4°C or -20°C as instructed and should not be used past the stated expiration date.
