Human Myosin Heavy Chain 3, Skeletal Muscle, Embryonic ELISA Kit

This sandwich ELISA kit from Antibodies.com enables in vitro quantitative determination of human Myosin Heavy Chain 3, Skeletal Muscle, Embryonic (Heavy Chain Myosin/MYH3) in tissue homogenates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate. Standards and samples are added to wells containing immobilized capture antibody; bound target is detected sequentially using a Biotinylated Anti-Heavy Chain Myosin/MYH3 antibody, an HRP-Avidin conjugate, and TMB substrate. The reaction is stopped with acidic stop solution and absorbance is read at 450 nm on a colorimetric microplate reader, with target concentration calculated against the standard curve.

Assay performance includes a sensitivity of <0.126 ng/ml and a working range of 0.312-20 ng/ml, with a total assay time of 4h 30m. Validation data shows spike-and-recovery averages of 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%), and 84% in heparin plasma (range 80-89%) across n=5 samples per matrix. Linearity upon dilution from 1:2 to 1:16 was demonstrated in serum (74-107%), EDTA plasma (79-108%), and heparin plasma (82-105%).

Kit components include a pre-coated 96-well microplate (12 x 8-well strips), lyophilized standard, biotinylated detection antibody, HRP-avidin conjugate, wash buffer (30X concentrate), sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, with component-specific storage at +4°C or -20°C as instructed.
