Human Myosin Heavy Chain 1, Skeletal Muscle, Adult ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Myosin Heavy Chain 1 (MYH1). The kit is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and reactivity is confirmed for human samples.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Myosin Heavy Chain 1/MYH1 is immobilised on the plate; standards and samples are added and the bound analyte is detected using a Biotinylated Anti-Myosin Heavy Chain 1/MYH1 antibody followed by an HRP-Avidin conjugate. TMB substrate is used to visualise the reaction, producing a yellow product after addition of the acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. Total assay time is approximately 4 hours 30 minutes. Kit components include the pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, stored at +4°C or -20°C as indicated.

Manufacturer-reported analytical specifications include a measuring range of 0.156–10 ng/ml and a sensitivity of <0.059 ng/ml. Reported spike-and-recovery values are: serum (n=5, range 80–102%, average 91%), EDTA plasma (n=5, range 81–100%, average 90%) and heparin plasma (n=5, range 80–89%, average 84%). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions, with serum ranging 87–107%, EDTA plasma 79–108% and heparin plasma 82–105% across the dilutions tested.
