From Antibodies.com, the Human Myoglobin ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of human Myoglobin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay is read on a colorimetric microplate reader at 450 nm, with a product range of 0.781–50 ng/ml and a sensitivity of <0.30 ng/ml. Total assay time is 4 hours 30 minutes.

The assay uses a 96-well microtiter plate pre-coated with a Myoglobin-specific capture antibody. Standards and samples are added to the wells and bound Myoglobin is detected using a biotinylated anti-Myoglobin antibody followed by an HRP-avidin conjugate. The HRP enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that turns yellow after addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of Myoglobin captured in each well, and the concentration is calculated by reference to a standard curve.

Reported sample recovery ranges from 80–102% (average 91%, n=5) in serum, 81–100% (average 90%, n=5) in EDTA plasma, and 80–89% (average 84%, n=5) in heparin plasma. The kit is supplied with a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or –20°C as instructed in the protocol manual, and the kit should not be used past its expiration date.
