Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Myeloperoxidase in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the quantitative sandwich enzyme immunoassay technique, in which an antibody specific for Myeloperoxidase has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added into the wells, and the Myeloperoxidase present is bound by the immobilized antibody.

Following incubation and washing, a Biotinylated Anti-Myeloperoxidase Antibody binds the captured analyte, after which an HRP-Avidin conjugate is added and incubated. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Myeloperoxidase captured in each well. Concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve. Total assay time is 4 hours 30 minutes on a pre-coated microplate platform of 12 x 8 well strips, read on a colourmetric microplate reader.

The assay has a reported product range of 0.781-50 ng/ml and a sensitivity of 0.33 ng/ml. Reported recovery values are 80-102% in serum (average 91%, n=5), 81-100% in EDTA plasma (average 90%, n=5), and 80-89% in heparin plasma (average 84%, n=5). Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard (2 vials), detection solution A (120 µl), detection solution B (120 µl), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers (5 adhesive strips). Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
